After high school, Nele Mueller-Stöfen graduated as an actress from school of music and acting in Graz. First engagement at theatre in Münster and Düsseldorf. 1993 she got an award for best young actress as "Gretchen" in the play "Faust".
Again and again we see her on stages like Kampnagel in Hamburg, the Renaissancetheatre and Sophiensäle in Berlin.
In 1994 Mueller-Stöfen made her film debut under director Rainer Kaufmann in "Einer meiner ältesten Freunde". It followed movies like "Dunkel" (1999),
"Commercial Man"(2001) made by Lars Kraume, "Gierig" by Oskar Röhler, "Jimmy the Kid" (1997) by Wolfgang Diekmann, "Frau2 sucht Happy End" (1998) by Edward Berger, „Das Herz ist ein dunkler Wald“ (2006) by Nicolette Krebitz, "Der Herrscher von Edessa"( 2007) by Tim Trachte and "Jack" (2013) by Edward Berger.
Various leading roles also in television films. Inter alia in "Buddies" (1997) by Roland Suso Richter, "Die Beischlafdiebin" (1998) von Christian Petzold, "Das Finale" by Sigi Rothemund "Ein tödliches Wochenende" (2001) by Torsten C. Fischer, "Der Solist" ( 2003) by Stefan Wagner, "Das Staatsgeheimnis" (2002), which was awarded with the 3Sat Public Price, by Mattias Glasner, "Einer bleibt sitzen" (2008) by Tim Trageser and "Familienfeste" (2014) by Lars Kraume.
She is writing scripts since 2010. Together with the director Edward Berger she wrote the script „Jack“, which was shown in competition at the 2014 Berlinale. She also wrote the script for her short film AM STRAND which she will direct this year.
